Preclinical Pharmacology Protocols

Preclinical Rodent Dosing & In Vivo Administration Protocols

Standards for dose-volume formulation, route selection, and ethical animal welfare limits in cancer research.

Accurate injection volume calculation is a foundational prerequisite for preclinical drug discovery, pharmacokinetic (PK) profiling, and xenograft tumor efficacy studies. The universal formula V = (Dose ร— BW) / C ensures individual animals receive precise weight-adjusted milligram doses.

Route-Specific Guidelines for Mice (Mus musculus)

Diehl et al. (2001) define standard maximal safe bolus volumes for adult mice: Intraperitoneal (IP): โ‰ค 20 mL/kg (ideal 10 mL/kg); Oral gavage (PO): โ‰ค 10 mL/kg (max 20 mL/kg with vehicle caution); Subcutaneous (SC): โ‰ค 10 mL/kg (split over multiple sites if viscous); Intravenous (IV): โ‰ค 5 mL/kg bolus (or up to 10 mL/kg slow infusion).

Salt Form & Molecular Weight (RMM) Corrections

When preparing doses from salt compounds (e.g. hydrochloride ยทHCl, mesylate, or sodium salts), investigators must correct for the active free-base fraction: Weighing Factor = RMM(salt) / RMM(free base). Forgetting this correction causes systematic underdosing by 10% to 35%.
